研究DNA/RNA 與聚陽(yáng)離子的靜態(tài)和動(dòng)態(tài)復(fù)合行為對(duì)于探討聚電解質(zhì)的相互作用機(jī)理,開(kāi)發(fā)高效基因傳遞載體,以及研究原始細(xì)胞的動(dòng)態(tài)模型都有重要意義。在靜態(tài)復(fù)合中,我們重點(diǎn)探討了DNA復(fù)合物的長(zhǎng)程動(dòng)力學(xué)行為,發(fā)現(xiàn)歧化是影響復(fù)合物動(dòng)力學(xué)的主要因素。通過(guò)調(diào)控歧化速率,構(gòu)筑了動(dòng)力學(xué)控制的膠囊結(jié)構(gòu)。在動(dòng)態(tài)復(fù)合中,首先模擬血液流動(dòng)引起的剪切作用,探討了外加剪切力對(duì)DNA復(fù)合物穩(wěn)定性的影響,并據(jù)此開(kāi)發(fā)了具有肺靶向功能的siRNA傳遞載體。 接下來(lái),利用電場(chǎng)來(lái)調(diào)控DNA與聚陽(yáng)離子在非平衡態(tài)的復(fù)合行為。結(jié)果表明,二者形成的復(fù)合物發(fā)生了周期性的內(nèi)分區(qū)化,尺寸和形狀的動(dòng)態(tài)漲落,混沌式的增長(zhǎng)和融合等許多與生命活動(dòng)類似的行為。在周期性的內(nèi)分區(qū)化過(guò)程中還伴隨著物質(zhì)的運(yùn)輸,主要表現(xiàn)為聚陽(yáng)離子含量的減少。我們用電場(chǎng)力學(xué)引起的聚電解質(zhì)復(fù)合物的解散,繼而引起的滲透壓變化來(lái)解釋內(nèi)分區(qū)化行為。這些結(jié)果都表明在非平衡態(tài)下研究復(fù)合物粒子的動(dòng)態(tài)行為對(duì)于開(kāi)發(fā)原始細(xì)胞,探索生命起源具有指導(dǎo)意義。
