報告摘要:

研究DNA/RNA 與聚陽離子的靜態和動態復合行為對于探討聚電解質的相互作用機理,開發高效基因傳遞載體,以及研究原始細胞的動態模型都有重要意義。在靜態復合中,我們重點探討了DNA復合物的長程動力學行為,發現歧化是影響復合物動力學的主要因素。通過調控歧化速率,構筑了動力學控制的膠囊結構。在動態復合中,首先模擬血液流動引起的剪切作用,探討了外加剪切力對DNA復合物穩定性的影響,并據此開發了具有肺靶向功能的siRNA傳遞載體。 接下來,利用電場來調控DNA與聚陽離子在非平衡態的復合行為。結果表明,二者形成的復合物發生了周期性的內分區化,尺寸和形狀的動態漲落,混沌式的增長和融合等許多與生命活動類似的行為。在周期性的內分區化過程中還伴隨著物質的運輸,主要表現為聚陽離子含量的減少。我們用電場力學引起的聚電解質復合物的解散,繼而引起的滲透壓變化來解釋內分區化行為。這些結果都表明在非平衡態下研究復合物粒子的動態行為對于開發原始細胞,探索生命起源具有指導意義。
