摘要:目的 探討多功能納米材料CDPF和CDP運載AEG-1 siRNA進入骨肉瘤細胞能力以及其對細胞增殖、侵襲能力的影響。方法 通過體外細胞瞬時轉(zhuǎn)染實驗將CDPF/AEG-1 siRNA或CDP/AEG-1 siRNA運載入143B細胞,用Western-blot檢測AEG-1 蛋白水平的變化,分別用MTT法和侵襲實驗檢測轉(zhuǎn)染復(fù)合物對143B細胞增殖能力和侵襲能力的影響。結(jié)果 CDPF和CDP成功的將AEG-1 siRNA運載至143B細胞,與對照組比較,AEG-1蛋白水平顯著降低,下調(diào)AEG-明顯抑制143B細胞的增殖和侵襲能力,且CDPF比CDP具有更好的優(yōu)勢。結(jié)論 CDP可作為有效的新型基因轉(zhuǎn)染載體,葉酸的接入可提高其效率。
